I will be on a business travel to Hefei next week. I’ve only been to Hefei a couple of times in the past few years either by flight or train. Both were not so good. Flights were not so frequent and trains took too much time. I knew that now bullet trains runs between Shanghai and Hefei from last year and the journey only takes about 3.5 hours. So I think it is the best way to travel Shanghai to Hefei. First class seats cost about 185RMB and second class cost about 150RMB. Time schedule as below, hope it might be useful for others.

Please note all the bullet trains departure from Shanghai Railway Station not South one.
